Gebraucht, aber gut erhalten. - The better understanding of neuron-glia interactions and their influence on synaptic neurotransmission in the prefrontal cortex (PFC) is crucial to address important issues related to the implication of these interactions for emotional, cognitive and mnemonic processes, information transactions and short-term memory. The main neuronal population of the PFC includes the pyramidal cells, which are anatomically distributed among the different cortical layers. In the rat, layer V pyramidal cells send the main efferent projections of the PFC and were previously described to be endowed with group I metabotropic glutamate receptors (mGluR) which can selectively modulate NMDA-type glutamate receptors. This effect was described to occur after astrocytic-release of glutamate, triggered by the endogenous P2 receptor agonist ATP. The goal of the first part of this work was to assess whether synaptic glutamate was responsible for this effect, and to study the implications of astrocytic glutamate uptake impairment on the postsynaptic glutamatergic activation. We used the patch-clamp technique to record the activity of pyramidal neurons and in addition a whole palette of pharmacological tools, including glutamate uptake and mGluR inhibitors, to access whether synaptic glutamate would be responsible for a similar effect.
